Guardian

Support heroes which use warding magic to protect and empower allies in battle.  A good complement to a wide variety of fighting styles.

Guardians don't have direct offensive or healing skills, but they have the ability to make all of their allies more effective with well-placed buffs and protection spells.  The Guardian's support skills are useful for a variety of heroes.

Protect
Increases the armor of a friendly unit for 20 seconds.

Protect Information
Casting Protect on any allies taking damage can go a good ways towards keeping them alive, and can dramatically reduce the amount of healing you need.

Because of the way armor works, Protect will be the most help (proportionately) to characters with lower-grade armor.

 

Blessed Vigor
Temporarily increases the strength of a friendly hero.

Blessed Vigor Information
Blessed Vigor can keep someone alive longer by increasing their maximum health and health regeneration, but it is most useful for increasing the damage of strength heroes.  +50 strength provides +50 damage if strength is your primary attribute, and that can be a huge boost.

 

Blessed Dexterity
Temporarily increases the agility of a friendly hero.

Blessed Dexterity Information
Blessed Dexterity increases the armor and attack rate of a hero, and also increases the attack damage of agility heroes.  This is a good buff to a front-line hero's effectiveness, especially on agility heroes.

 

Blessed Insight
Temporarily increases the intelligence of a friendly hero.

Blessed Insight Information
Blessed Insight will increase the maximum mana and mana regeneration of a hero, which can be quite helpful to have at the start of a big fight, and can also allow you to cast high-mana-cost spells which would otherwise be very difficult to cast.  The bonus intelligence also increases the attack damage for intelligence heroes, which can be a substantial improvement to their offensive capabilities.

 

Invincibility
Makes a friendly unit temporarily invulnerable.

Invincibility Information
You can use Invincibility to save an ally who's about to die, or to force monsters to choose a different target than the one they're currently attacking.  Keep in mind that monsters with no valid targets to attack will tend to move past you towards the tower, so even if you've got the mana for perpetual invincibility, it may not be a good idea.

Mystic Wall Information
When you're taking a lot of damage, Mystic Wall may be more efficient than healing or other methods of damage prevention, and it can help you stay alive a lot longer without requiring you to cast anything.  However, it drains a substantial amount of mana, so be sure to turn it off when you don't need it.

Reduces damage taken.

Fortitude Information
Fortitude will prolong your life significantly by reducing all forms of damage received.  This is a useful mastery for all heroes, but especially those who tend to take the most damage from monsters.

 

Mobility

Mobility Information
Mobility allows you and anyone near you to move around faster, which can be useful for closing on ranged enemies, running away from monsters when injured, and getting around your base between waves.  The range of this aura is fairly high, so you don't need to be especially close to confer bonuses on your allies.  Mobility doesn't grant a direct combat advantage, but it's inexpensive to learn, and it can help you use your time more efficiently.

Mobility stacks with Endurance and other speed-enhancing abilities.
